Preparation Method
Sample support involves surrounding fibre bundles with liquid polymers that harden into a rigid block for microtome slicing. Using acrylic resin embedding ensures that the internal structures of flax bundles remain undistorted during the mechanical cutting of thin sections. The polymer fills the interstitial spaces between individual fibres and stabilizes the fragile middle lamella.
Microtomy Benefit
Hardness of the resulting block determines the achievable thickness of the sample slices. Because the acrylic resin embedding creates a uniform density across the mounting medium, the diamond or glass knives used in Chinese mill laboratories produce sections measuring only a few microns. Such precision allows for the clear identification of individual cell walls under high magnification.
This uniformity prevents the crushing or tearing of the lumen during the preparation of export quality documentation and ensures that the internal anatomy of the flax remains representative of the bulk shipment.
Application Boundary
Stability provided by this technique stops at the point of chemical interaction with specific staining reagents. While acrylic resin embedding works for light microscopy, certain solvent-heavy dyes may soften the matrix or fail to penetrate the cured plastic. The method is used primarily for morphology checks rather than bulk tensile testing.
